ABSTRACT:
Utilization of TRU-ADhanCe™ Technology to Significantly Enhance the ADCC Potency of Immunoglobulin Fc-Bearing Molecules
TRU-ADhanCe™ is a broadly applicable manufacturing technology designed to markedly augment the antibody-dependent cellular cytotoxicity (ADCC) of active Fc-containing molecules. With the simple addition of the glycosidase inhibitor castanospermine during the cell culture stage, core-fucose of Fc glycans is significantly reduced resulting in enhanced ADCC activity, while protein production is maintained at high level. This presentation will provide an overview of the technology and a case study of its application to a Small Modular Immunopharmaceutical (SMIP)™ molecule.

Beyond SMIPs™: The Development of Small Multi-Specific Immunotherapeutics
Engineering specificity for more than one therapeutic target into antibodies has been challenging. We have developed several technologies, including the Scorpion™ platform, to bind more than one target simultaneously, whether soluble ligands or cell surface receptors. These scaffolds retain the advantages of antibodies - high affinity, lack of immunogenicity, long plasma halflife - and are amenable to large-scale production and purification.

TRU-016 Phase 1 and preclinical data will be presented at the 2009 American Society of Hematology (ASH) Annual Meeting Dec. 5-8, 2009 in New Orleans, Louisiana. The presentations will include additional positive data from 1) an ongoing Phase 1 clinical trial of TRU-016 for the treatment of chronic lymphocytic leukemia (CLL), and 2) a preclinical study demonstrating the effect of Trubion's Tru-ADhanCe™ technology in promoting enhanced natural killer cell mediated cytotoxicity against primary CLL cells. TRU-016 is Trubion's novel CD37-targeted therapy for the treatment of B-cell malignancies and is being developed in collaboration with Facet Biotech Corporation.

Arthritis, lupus, gout and other debilitating inflammatory conditions are chronic autoimmune illnesses characterized by an immune system that has become misdirected. An estimated 43 million people in the United States alone develop autoimmune rheumatic conditions. By 2020, there will be 60 million diagnosed. Unfortunately, many patients do not fully respond to available treatments and those treatments that are available are known to cause further complications, such as PML. With the discovery of new molecular targets and pathways, companies are advancing new classes of medicines for autoimmune inflammatory diseases, including oral kinase inhibitors in RA and monoclonal antibodies in lupus. The current pipeline may hold the answer for millions of patients. Companies with late-stage compounds for arthritis and inflammatory diseases, key opinion leaders and advocates will provide insights on experimental new autoimmune therapies, clinical need and application and the development landscape.
